The best spot to have the code fro the check is anywhere that alignment 
is changed.  If you can only change alignment by killing, then only have 
it where your alignment changes due to a kill.  Having it check every 
player every second is, to put it bluntly, a serious waste of processor 
time, UNLESS, you have another way to change alignment, and if you do, 
just put another check wherever alignment can be changed for the 
positive.  (ie. YOu don't need to put a check in spell_energy_drain as it 
ALWAYS lowers alignment)
